Adherent Cell Transfection

Adherent transfection is generally more forgiving than suspension transfection, but confluency, attachment strength, and monolayer uniformity all directly affect efficiency in ways that are easy to overlook.

Confluency Sensitivity:

Most adherent lines have a fairly narrow confluency window (commonly 60–80%) where transfection efficiency peaks. Under-confluent cells are often unhealthy or slow-growing, while over-confluent cells have reduced surface area per cell and lower division rates, both of which lower uptake and expression.

Detachment During Media Changes and Washing:

Loosely adherent lines (HEK293 is a well-known example) can lift off the plate during media changes, complex addition, or washing steps. Cell loss during these steps not only reduces total yield but also skews results if detachment is uneven across the plate.

Uneven Complex Distribution Across the Monolayer:

Transfection complexes settle by gravity onto the cell layer, which means uneven pipetting, plate rocking, or well geometry can create regions of higher and lower cargo exposure across a single well, especially in larger-format plates.

Cell-Cell Contact and Morphology-Dependent Uptake:

As adherent cells approach confluency, increased cell-cell contact and monolayer crowding can restrict membrane accessibility and reduce endocytosis relative to more sparsely plated, well-spread cells.

Extended Adhesion-Related Stress:

Some primary and slow-adhering cell types spend a meaningful amount of time re-establishing attachment after passaging before they are transfection-ready, and transfecting too soon after plating can compound attachment stress with transfection-related toxicity.
