Transfection of primary cells, such as Human Chondrocyte (HC, NHAC, HC-RA, HC-OA), has always been challenging, both technically and financially. The most common systems currently used for relatively efficient molecular delivery to these cells are expensive electroporation instruments. However, electroporation usually leads to cell death, primarily when the electrical fields cause permanent permeabilization of the membrane and the consequent loss of cell homeostasis, in a process known as irreversible electroporation.

Human Chondrocyte (HC, NHAC, HC-RA, HC-OA) Avalanche® Transfection Reagent is a new, proprietary solution specifically designed for transfection on HC. Cell Name: Human Chondrocyte (HC, NHAC, HC-RA, HC-OA)

Cell Synonyms: HC, NHAC, HC-RA, HC-OA

Chondrocytes produce and maintain the extracellular matrix of cartilage. Human Chondrocytes are usually derived from normal human articular cartilage. Human Chondrocytes growing in serial monolayer culture lose their phenotypic markers and became de-differentiated. This de-differentiation stage can be reversed by culturing them in semi-solid gel. Human Chondrocyte has served as a useful model for studying chondrocyte differentiation and the effects of cytokines and growth factors on the suppression and maintenance of differentiated cartilage phenotype.

Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
