Suspension Cell Transfection Reagents
Suspension cells grow freely in liquid culture instead of attaching to a surface, which makes them the format of choice for large-scale bioreactor production but also changes almost everything about how transfection reagents need to behave. Compared to adherent formats, suspension transfection has to work around continuous agitation, no fixed surface for complex settling, and, in many cases, fully serum-free, chemically defined media.

The Challenges of Suspension Cell Transfection
Suspension transfection is not simply “adherent transfection without the plate.” The absence of a surface changes the physics of complex formation, delivery, and mixing, which means suspension-specific optimization is usually required rather than optional.
No Surface for Complex Settling:
In adherent culture, lipoplexes or polyplexes settle by gravity directly onto the monolayer, concentrating cargo near the cell surface. In suspension, cells and complexes are both moving through the same volume, so uptake depends on random collision rather than settling, which typically lowers effective local concentration.
Shear Stress from Agitation:
Suspension cultures are kept in motion by shaking, stirring, or rocking platforms. This constant agitation can disrupt fragile lipid/nucleic acid complexes before they’re taken up, and can also stress the cells themselves, compounding any transfection-related toxicity.
Serum-Free and Chemically Defined Media Compatibility:
Most industrial and many research suspension lines (CHO, HEK293) are adapted to serum-free, chemically defined media. Many transfection reagents optimized for serum-containing adherent protocols lose efficiency or become toxic in these formulations, so reagents need to be validated specifically in serum-free suspension conditions.
Complex Formation Timing at Scale:
Adherent protocols are often forgiving of small timing variations because volumes are small. In suspension, especially at bioreactor scale, complex formation, addition, and mixing need to be timed and scaled carefully, since uneven addition across a large culture volume can create pockets of over- or under-dosed cells.
Aggregation and Clumping:
Some suspension-adapted lines are prone to cell clumping, which can trap complexes at the periphery of an aggregate and prevent even access to cells in the center, reducing overall population-level efficiency even when total DNA uptake looks adequate.

Suspension vs. Adherent Transfection
Switching a cell line between suspension and adherent formats is possible for some lines but slow, and the resulting suspension-adapted cells often behave differently from their adherent counterparts including in transfection efficiency.
As a general rule, suspension formats favor reagents with strong colloidal stability (complexes that hold together under agitation) over reagents optimized purely for maximum peak efficiency in a static, serum-containing environment.
