Transfection of primary cells, such as Human Bladder Epithelial Cell (HBlEpC), has always been challenging, both technically and financially. The most common systems currently used for relatively efficient molecular delivery to these cells are expensive electroporation instruments. However, electroporation usually leads to cell death, primarily when the electrical fields cause permanent permeabilization of the membrane and the consequent loss of cell homeostasis, in a process known as irreversible electroporation.

Human Bladder Epithelial Cell (HBlEpC) Avalanche® Transfection Reagent is a new, proprietary solution specifically designed for transfection on HBlEpC. Beyond the role of the bladder epithelium in forming a barrier, this tissue may play an unappreciated but important role in water, salt, and solute homeostasis. One of the most exciting functions of the bladder epithelium is its potential role as a sensory transducer.

Urothelial carcinoma of the bladder is unique among epithelial carcinomas in its divergent pathways of tumorigenesis. Low-grade papillary tumors rarely become muscle-invasive and they frequently harbour gene mutations that constitutively activate the receptor tyrosine kinase®Ras pathway. By contrast, most high-grade invasive tumors progress to life-threatening metastases and have defects in the p53 and the retinoblastoma protein pathways.

Human bladder epithelial cells (HBlEpC) can be used as a powerful tool in studying not only bladder epithelial development and function, but also the processes by which epithelial cells achieve and maintain their specialized plasma membrane domains, and how these functions are disrupted in disease.

Transfection Reagent Features:

This test result was confirmed with repeat experiments. The candidate formulas were also tested in human bladder epithelial cell (HBlEpC) by tranfecting GFP expression plasmid, and got the same high transfection efficiency after flow cytometry analysis. The one that showed the optimal balance of potent & low cytotoxicity (percentage of 7AAD positive cells) among those candidate formulas was later named as Human Bladder Epithelial Cell (HBlEpC) Avalanche Transfection Reagent.

Recommended protocols for these cells will be provided with the reagent. The protocols usually provide satisfactory transfection efficiency with invisible cytotoxicity. However, optimization may be needed for certain type of cells. Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
