Transfection of primary cells, such as Human Epidermal Melanocyte (HEM, NHEM), has always been challenging, both technically and financially. The most common systems currently used for relatively efficient molecular delivery to these cells are expensive electroporation instruments. However, electroporation usually leads to cell death, primarily when the electrical fields cause permanent permeabilization of the membrane and the consequent loss of cell homeostasis, in a process known as irreversible electroporation.

Human Epidermal Melanocyte (HEM, NHEM) Avalanche® TRansfection Reagent is a new, proprietary solution specifically designed for transfection on HEM. The proprietary formulation of lipids and polymers ensures the highest possible transfection efficiencies and viabilities for HEM. Cell Synonyms: HEM, NHEM, NHEM-Ad, NHEM-Neo

Human Epidermal Melanocytes (HEM) are the pigment producing cells located in the epidermal layers of all mammalian skin. The function of the melanocyte is to produce melanin, the pigment that gives skin its color. Melanin protects skin from UV damage.

Melanocytes in cosmetic studies are used to test:

  • Skin lightening compounds for moles-freckles-nevi-age spots
  • Skin darkening products – tan enhancing products
  • UV protection compounds – sunscreen

Melanocyte in medical research often centers on melanoma as well as various dermatological diseases or related disorders like:

  • Albinism and vitiligo
  • Hormonal treatments
  • Thyroid conditions
  • Psoriasis
  • Burns
  • Scars

The Transfection Reagent Features:

  • Broad spectrum for the transfection of large plasmid, mRNA, siRNA, and/or other type of nucleic acids, which is best for co-transfection of different type and/or size of nucleic acids.
  • Specifically optimized to deliver plasmid DNA into HEM

Recommended protocols for these cells will be provided with the reagent. The protocols usually provide satisfactory transfection efficiency with invisible cytotoxicity. However, optimization may be needed for certain type of cells. Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
