Transfection of primary cells, such as PrSc, has always been challenging, both technically and financially. The most common systems currently used for relatively efficient molecular delivery to these cells are expensive electroporation instruments. However, electroporation usually leads to cell death, primarily when the electrical fields cause permanent permeabilization of the membrane and the consequent loss of cell homeostasis, in a process known as irreversible electroporation.

Human Prostate Stromal Cell (PrSC) Avalanche® Transfection Reagent is a new, proprietary solution specifically designed for transfection on PrSc. The proprietary formulation of lipids and polymers ensures the highest possible transfection efficiencies and viabilities for PrSc. Prostate cells provide a glandular function in the body by generating fluid which serves several functions in reproduction. Prostates consist of glandular epithelium embedded in a fibro-muscular stroma. The secretory luminal layer of the epithelium is made up of tall columnar cells that are responsible for the production of PSA, PAP and human kallikrein-2 that are secreted as part of the seminal fluid into the glandular lumina.

Human Prostate Stromal Cells (PrSC) are necessary for the normal growth and survival of epithelial cells in the prostate. A number of findings have suggested interactions between PrSC and the epithelium as well as importance of the extracellular matrix in prostate development and differentiation; alterations of these interactions may lead to prostate cancer progression (Djakiew et al., 1990; Cunha et al., 1996, 2002). Stromal®epithelial interactions appear to be reciprocal, mediated by steroid hormones, soluble and insoluble factors and their receptors, and constituents of the extracellular matrix (Chung, 1993; Rowley, 1999).

PrSC can be used for the following applications:

Transfection reagent features:

Recommended protocols for these cells will be provided with the reagent. The protocols usually provide satisfactory transfection efficiency with invisible cytotoxicity. However, optimization may be needed for certain type of cells. Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
