HepG2/C3A Cell Avalanche® Transfection Reagent is a new class of unique chemical formulations specifically formulated and optimized for transfecting HepG2/C3A cells. Comments: C3A is clonal derivative of Hep G2 that was selected for strong contact inhibition of growth, high albumin production, high production of alpha fetoprotein (AFP) and ability to grow in glucose deficient medium. As the cells become confluent, there is a marked reduction in AFP secretion and an increase in albumin secretion. Gluconeogenesis activity is strongly oxgen dependent. The cells have nitrogen metabolizing activity comparable to perfused rat livers.

The candidate formulas were also tested on HepG2/C3A cells by transfecting GFP expression plasmid, and got the same high transfection efficiency after flow cytometry analysis. The one that showed the optimal balance of potent & low cytotoxicity (percentage of 7AAD positive cells) among those candidate formulas was later named as HepG2/C3A Cell Avalanche Transfection Reagent.

Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
