ANXA5 (Annexin A5), Human
Catalog Number: EZC-AN5-H1
Synonym: ANXA5, ANXA-5, ANX5, ANX-5, ENX2, ENX-2, PP4, PP-4, annexin V, Annexin A5
Source: Recombinant human Anexin A5 (rhANXA5) Met1-Aps320 (Accession # AAH01429) was produced in E.coli cells.
Molecular Characterization: rhANXA5 contains C-terminal polyhistidine tag and has a calculated MW of 36.8 kDa. DTT-reduced protein migrates as 36.8 kDa estimated polypeptide in SDS-PAGE.
Purity: >98% as determined by SDS-PAGE. All lots are greater than 97% pure.
Endotoxin: Less than 1.0 EU per g of the rhANXA5 by the LAL method.
Formulation: Bulk protein in a 0.22 m filtered solution of PBS, pH 7.4 and delivered as liquid formulation or lyophilized powder. Normally 5-8% trehalose and mannitol are added as protectants before lyophilization.
Storage: Store at -20? in lyophilized state after receipt. For long term storage, upon reconstitution rhANXA5 should be aliquot and store at -20? or -80?. Avoid repeated freeze-thaw cycles.
Background: Anexin A5 is a phospholipid binding protein, which binds with high affinity and selectivity to PS in the presence of calcium. PS is predominantly located in membrane leaflets, which face the cytosol. However, recent findings show that each cell type has the molecular machinery to expose PS at its cell surface. This machinery is activated during the execution of apoptosis. Once PS is exposed at the cell surface it exhibits procoagulant and proinflammatory activities. Anexin A5 will bind to the PS-exposing apoptotic cell and can inhibit the procoagulant and proinflammatory activities of the dying cell. Anexin A5 has also been identified as an anticoagulant protein in the blood coagulation cascade, by acting as an inhibitor of prothrombin activation. The presence of antibodies to Anexin A5 is associated with systemic lupus erythematosus (SLE), recurrent spontaneous abortions and systemic sclerosis (SSc). (1,2)
