Source: Recombinant human ANGPT2 (rhANGPT2) Met1-Phe496 (Accession # AAI26201.1) fused with 6his tag at C-terminal was produced in human HEK 293 cells.
Molecular Characterization: rhANGPT2 is fused with a 6his tag at C-terminal and the protein has a calculated MW of 50.1kDa. The predicted N-terminal is Asp68 & Tyr19. DTT-reduced protein migrates as 66-90 kDa in SDS-PAGE due to glycosylation.
Purity: >90% as determined by SDS-PAGE. All lots are greater than 90% pure.
Endotoxin: Less than 2.0 EU per 1 g of the rhANGPT2 by the LAL method.
Formulation: Bulk protein in a 0.22 m filtered solution of PBS, pH 7.4 and delivered as liquid formulation or lyophilized powder. Normally 5-8% trehalose and mannitol are added as protectants before lyophilization.
Storage: Store at -20? in lyophilized state after receipt. For long term storage, upon reconstitution rhANGPT2 should be aliquot and store at -20? or -80?. Avoid repeated freeze-thaw cycles.
Background: Angiopoietin-2, also known as ANGPT2, AGPT2, ANG2, and is a secreted glycoprotein that plays a complex role in angiogenesis and inflammation [1, 2]. Ang2 is widely expressed during development, but it is restricted postnatally to highly angiogenic tissues such as the placenta, ovaries, and uterus [3]. It is particularly abundant in vascular endothelial cells (EC) where it is stored in intracellular WeibelPalade bodies [1, 3, 4]. Both Ang2 and the related Angiopoietin1 (Ang1) are ligands for the receptor tyrosine kinase Tie 2 [2]. Ang2 functions as a proangiogenic factor, although it can also induce EC death and vessel regression [5, 6]. Upon its release from quiescent EC, it regulates vascular remodeling by promoting EC survival, proliferation, and migration and destabilizing the interaction between EC and perivascular cells [6]. Ang2 is required for postnatal vascular remodeling, and it cooperates with Ang1 during lymphatic vessel development [4]. It mediates the upregulation of ICAM1 and VCAM1 on EC, which facilitates the adhesion of leukocytes during inflammation. Ang2 competitively inhibit Ang1-induced endothelial cell responses mediated by Tie2, and reduces vascular integrity. But the role of Ang2 is controversial since the opposite outcomes has been reported in other studies. Over-expression of Ang2 disrupts the vascular remodeling, induce endothelial cell apoptosis, and may play an important regulating role in tumor angiogenesis. Ang2 also promotes the neuronal differentiation and migration of subventricular zone progenitor cells [7]
