Molecular Characterization: rhALK3, a 141 amino acids protein with polyhistidine tag at C-terminus, and has a calculated MW of 15.7 kDa. The predicted N-terminal is Gln24. DTT-reduced protein migrates as 26 and 32 kDa protein due to different glycosylation.
Purity: >97% purity as determined by SDS-PAGE of reduced rhALK3.
Endotoxin: Less than 0.01 ng endotoxin per g rhALK3.
Formulation: Bulk protein in a 0.22 m filtered solution of PBS, pH 7.4 and delivered as liquid formulation or lyophilized powder. Normally 5-8% trehalose and mannitol are added as protectants before lyophilization.
Storage: Store at -20? in lyophilized state after receipt. For long term storage, upon reconstitution rhALK3 should be aliquot and store at -20? or -80?. Avoid repeated freeze-thaw cycles.
Background: The bone morphogenetic protein receptor, type IA also known as BMPR1A or ALK3 is a protein which in humans is encoded by the BMPR1A gene. BMPR1A has also been designated as CD292 (cluster of differentiation 290). The bone morphogenetic protein (BMP) receptors are a family of transmembrane serine/threonine kinases that include the type I receptors BMPR1A (this protein) and BMPR1B and the type II receptor BMPR2. These receptors are also closely related to the activin receptors, ACVR1 and ACVR2(1). Alk3-mediated BMP signaling in AV endocardial/mesenchymal cells plays a central role during cushion morphogenesis(2).
