Source: Recombinant human BMP2 (rhBMP2) Gln283-Arg396 (Accession # NP_001191) was produced in E.coli cells.
Molecular Characterization: rhBMP2 contains no "Tags" and has a calculated MW of 13 kDa. DTT-reduced protein migrates as a 13 kDa polypeptide and the non-reduced cystine-linked homodimer migrates as a 24 kDa protein.
Purity: >92% as determined by SDS-PAGE of 6 g reduced (+) and non-reduced (-) rhBMP2. All lots are greater than 92% pure.
Biological Activity: The ED50 as determined by the cytolysis of MC3T3-E1 cells is <50 ng/ml, corresponding to a specific activity of 2.0104 IU/mg.
Endotoxin: Less than 1.0 EU per 1 g of the rhBMP2 by the LAL method.
Formulation: Supplied as a 0.22 m filtered solution in 0.1M acetic acid, pH2.0 and delivered as liquid formulation or lyophilized powder. Normally mannitol or trehalose are added as protectants before lyophilization. Reconstitution instruction or specific concentrations are included in the hardcopy of CoA along with the products.
Storage: No activity loss was observed after storage at 4-8? for 1 month or -20? for 1 year in lyophilized state. rhBMP2 should be aliquoted and stored at -80? in liquid formulation, for long term storage. Avoid repeated freeze-thaw cycles.
Background: Human bone morphogenetic protein 2 (BMP2) is also known as human BMP-2, BMP-2, BMP2, BMP 2, h-BMP-2, rh-BMP-2, recombinant human BMP-2, recombinant BMP-2, BMP, is a member of the BMP subgroup belonging to the TGF-b superfamily of structurally related signaling proteins. BMP-2 is a potent osteoinductive cytokine, capable of inducing bone and cartilage formation in association with osteoconductive carriers such as collagen and synthetic hydroxyapatite. In addition to its osteogenic activity, BMP-2 plays an important role in cardiac morphogenesis and is expressed in a variety of tissues including lung, spleen, brain, liver, prostate ovary and small intestine. As implied by its name, BMPs promote and regulate bone development, growth, remodeling and repair, in both prenatal development and postnatal growth of eye, heart, kidney, skin, and other tissues.(2,3) BMP2 has been demonstrated to potently induce osteoblast differentiation in a variety of cell types, and induce apoptosis in human myeloma cell lines as a novel function. rhBMP2 can be utilized in various therapeutic interventions such as bony defects, delayed union, non-union fractures and osteoporosis.(1) .
