bFGF (Basic Fibroblast Growth Factor), Human
Catalog Number: EZC-BFF-H1
Synonym: FGF2, FGF-2, BFGF, FGFB, FGF, FGF-, FGF-beta HBGF-2 Basic fibroblast growth factor
Source: Recombinant human bFGF (rh-bFGF) Pro143-Ser288 (Accession # NP_001997) was expressed in E. Coli.
Molecular Characterization: rh-bFGF contains no
Purity: >98% as determined by SDS-PAGE of 6 g reduced rh-bFGF. All lots are greater than 98% pure.
Biological Activity: Measured in a cell proliferation assay using BALB/c 3T3 mouse embryonic fibroblasts. The ED50 for this effect is typically 0.02 - 0.1 ng/mL.
Endotoxin: Less than 1.0 EU per 1 g of the rh-bFGF by the LAL method.
Formulation: Bulk protein in a 0.22 m filtered solution of PBS, pH 7.4 and delivered as liquid formulation or lyophilized powder. Normally 5-8% trehalose and mannitol are added as protectants before lyophilization.
Storage: Store at -20? in lyophilized state after receipt. For long term storage, upon reconstitution rh-bFGF should be aliquot and store at -20? or -80?. Avoid repeated freeze-thaw cycles.
Background: FGF basic is a member of the FGF family of at least 23 related mitogenic proteins which show 35-60% amino acid conservation. FGF acidic and basic, unlike the other members of the family, lack signal peptides and are apparently secreted by mechanisms other than the classical protein secretion pathway. FGF basic has been isolated from a number of sources, including neural tissue, pituitary, adrenal cortex, corpus luteum, and placenta. This factor contains four cysteine residues, but reduced FGF basic retains full biological activity, indicating that disulfide bonds are not required for this activity.[1] bFGF is a critical component of human embryonic stem cell culture medium; the growth factor is necessary for the cells to remain in an undifferentiated state, although the mechanisms by which it does this are poorly defined. It has been demonstrated to induce gremlin expression which in turn is known to inhibit the induction of differentiation by bone morphogenetic proteins.[2] It is necessary in mouse-feeder cell dependent culture systems, as well as in feeder and serum-free culture systems.[3].
